    Funder: Immune Tolerance Network

Due Dates: January 15, 2027 (Letter of Interest initial review) | Rolling (RFP remains open)

Summary: Supports phase I/II multicenter clinical trials testing therapies and biomarkers to improve immunosuppression withdrawal in adult liver transplant recipients.

Key Information: Proposals must include a testable mechanistic hypothesis and human dosing and safety evidence for the proposed intervention.

    Description

The Immune Tolerance Network (ITN) seeks short proposals for phase I/II multicenter interventional clinical trials of tolerance in adult liver transplant recipients. The goal is to increase successful immunosuppression withdrawal (ISW) from approximately 15% in carefully selected recipients to more than 40%. Proposed approaches may include pharmacologic therapies, cellular therapies, candidate biomarkers that predict successful ISW, or combinations of these approaches.

Supported by the National Institute of Allergy and Infectious Diseases at the NIH, ITN integrates mechanism-based research into clinical trials of immune tolerance. This call addresses the long-term risks of immunosuppression, including infection, malignancy, and metabolic side effects. Trials may study de novo liver transplant candidates or recipients, or long-term patients with functioning transplants. ITN anticipates proposals enrolling 10–50 patients and prioritizes clear, strong mechanistic hypotheses.
